pGL4.81[hRlucCP/Neo]质粒载体简介

 pGL4.81[hRlucCP/Neo]载体(a–d)编码荧光素酶报告基因hRlucCP(海肾荧光素酶)。该载体旨在实现高表达并减少异常转录。此外,该载体还含有哺乳动物细胞选择性标记新霉素抗性标记,其中转录因子结合位点的数量已被减少,且该标记适用于哺乳动物密码子使用已优化。pGL4载体经过工程设计,其共有调控序列少于pGL3载体以及一个经过密码子优化以适应哺乳动物表达的合成报告基因。
 
pGL4.81[hRlucCP/Neo]载体是一个没有启动子的基础载体。然而,它包含一个多克隆位点允许克隆所选的启动子。hRlucCP报告基因包含两个蛋白质去稳定序列,即hCL1和hPEST。hRlucCP编码的蛋白质对变化的响应速度更快,幅度更大其转录活性不如hRluc基因,后者更为稳定。
